PALMITOYLATION AND THE YEAST CASEIN KINASE YCK2byIRENE PAPANAYOTOUMay 2011Advisor: Dr. Nicholas G. DavisMajor: PharmacologyDegree: Doctor of Philosophy ! The Saccharomyces cerevisiae type I casein kinase Yck2 is tethered to thecytosolic face of the plasma membrane through palmitoylation of its two C-terminalcysteines. Yck2 palmitoylation is mediated by the Golgi-localized palmitoyl transferaseAkr1. Yck2 is a cytosolic protein and is presumably synthesized on cytosolic ribosomes. While the trafficking events following palmitoylation of Yck2 at the Golgi are wellcharacterized, the initial trafficking events after synthesis remain unknown. We sought toelucidate these early events of how Yck2 might travel to the Golgi-localized Akr1 after itis synthesized.! Our initial experimental analysis of Yck2 utilized glass bead cell lysates. The useof glass beads to mechanically disrupt yeast cells is used in a variety of applications, including for the analysis of native protein function (e.g. protein-protein interaction, enzyme assays, membrane fractionations). We describe a surprising case of proteindenaturation and aggregation that is induced by this lysis protocol. Interestingly, whenyeast cells are subjected to glass bead disruption, non-palmitoylated, cytosolic forms of118

Molecular Biology of the Cell #E03-09-0682 (revised) The Yeast Casein Kinase Yck3p is Palmitoylated, then Sorted to the Vacuolar Membrane with AP-3-Dependent Recognition of a YXXΦ Adaptin Sorting Signal
Our previous work found the two yeast plasma membrane-localized casein kinases Yck1p and Yck2p to be palmitoylated on C-terminal Cys-Cys sequences by the palmitoyl transferase Akr1p. The present work examines a third casein kinase, Yck3p which ends with the C-terminal sequence Cys-Cys-Cys-Cys-Phe-Cys-Cys-Cys. Yck3p is palmitoylated and localized to the vacuolar membrane. While the C-terminal cy...
متن کاملProtein palmitoylation by a family of DHHC protein S-acyltransferases.
Protein palmitoylation refers to the posttranslational addition of a 16 carbon fatty acid to the side chain of cysteine, forming a thioester linkage. This acyl modification is readily reversible, providing a potential regulatory mechanism to mediate protein-membrane interactions and subcellular trafficking of proteins. The mechanism that underlies the transfer of palmitate or other long-chain f...
متن کاملThe yeast kinase Yck2 has a tripartite palmitoylation signal
The yeast kinase Yck2 tethers to the cytoplasmic surface of the plasma membrane through dual palmitoylation of its C-terminal Cys-Cys dipeptide, mediated by the Golgi-localized palmitoyl-transferase Akr1. Here, the Yck2 palmitoylation signal is found to consist of three parts: 1) a 10-residue-long, conserved C-terminal peptide (CCTP) that includes the C-terminal Cys-Cys dipeptide; 2) the kinase...
متن کاملPrenylated isoforms of yeast casein kinase I, including the novel Yck3p, suppress the gcs1 blockage of cell proliferation from stationary phase.
The GCS1 gene of the budding yeast Saccharomyces cerevisiae mediate the resumption of cell proliferation from the starved, stationary-phase state. Here we identify yeast genes that, in increased dosages, overcome the growth defect of gcs1 delta mutant cells. Among these are YCK1 (CK12) and YCK2 (CKI1), encoding membrane-associated casein kinase I, and YCK3, encoding a novel casein kinase I isof...
متن کاملModel organisms lead the way to protein palmitoyltransferases.
The acylation of proteins with palmitate and related fatty acids has been known for over 30 years, but the molecular machinery that carries out palmitoylation has only recently emerged from studies in the model organisms Saccharomyces cerevisiae and Drosophila. Two classes of protein acyltransferases (PATs) have been proposed. In yeast, members of a family of integral membrane proteins harborin...
